• [gyrA43](ts)-- The gyrA allele was transduced in from E. coli K-12.
• [gyrA43](ts)-- has a high spontaneous reversion frequency. We suggest frequent verification of the Ts phenotype.
• thyA755-- Strains with this marker must have their medium supplemented with thymine (25ug/ml). This is true even for rich medium such as LB. Otherwise thymine starvation can lead to thymineless death.
• polA1(Am)-- normal levels exonuc.;premature term.of polymerase <1% WT level. MMS^S,UV^S, viable at all temps. Grindley MGG143
• = good miniprep dsDNA quality
• Am = amber(UAG) mutation
• strR = streptomycin resist.
• ts = Temperature Sensitive
