CGSC Strain#: 6972
Strain Designation: KNK453
Source of Strain: K.N. Kreuzer
Sex: F-
Chromosomal Markers: [gyrA43](ts)
,
thyA45,
polA0,
uvrA103,
deo-225Strain Comments: - [gyrA43](ts)-- The gyrA allele was transduced in from E. coli K-12.
- [gyrA43](ts)-- has a high spontaneous reversion frequency. We suggest frequent verification of the Ts phenotype.
- thyA45-- was called tlr-25 and thyR25, in conflict with the thyA25 allele. The Tlr (Thy low req.) phenotype is now known to result from two mutations, thyA--, deo-.
- thyA45-- Strains with this marker must have their medium supplemented with thymine (25ug/ml). This is true even for rich medium such as LB. Otherwise thymine starvation can lead to thymineless death.
- polA0-- The source of this allele is not yet known.
- deo-225-- is in deoB or deoC. In combination with a thyA- allele, it results in low Thy-requirement (Tlr) phenotype.
- deo-225 was formerly called thyA45
- deo-225 was formerly called thyR25
- deo-225 was formerly called tlr-25
- ts = Temperature Sensitive
